does it really make any noticable diffrence in sound/output in a ported box?

u wont notice an audible difference, but to the meter there is usually a slight gain.

rounded edges on the port? Wind noise will go down. not so important in a trunk, but in an open hatch like mine it makes a huge difference.

well he shouldnt be getting port noise in the first place if his box was built right
You could do it, but dont expect a huge gain in output. All its really gonna do is BARELY get you less resistance in airflow

Not only should you roundover your port edges, you should roundover the outside angles as much as possible. Try to eliminate ALL the sharp angles in your box and you'll see a gain on the meter. 1" pvc is good for doing that. fiberglass it in smooth.

another good trick of the trade but requires a bit more resin is stand the box on its corner and poour the resin in .

also if you have a thick baffle. router the inside edge of your speaker cutout, give you a slight gain as well

how much do you router out of say, a 3/4 or 5/8 mdf?

Probably wont see much fo a gain from a 3/4" baffle, but just use a roundover bit

Im fibreglassing a custom box right now, instead of a wood box design, and am even resining the inside, to make the wood really smooth for better air flow. I will post picks after Im done, but for now its a customers and they dont want it show just yet.
